Directed attenuation to enhance vaccine immunity

چکیده

Many viral infections can be prevented by immunizing with live, attenuated vaccines. Early methods of attenuation were hit-and-miss, now much improved genetic engineering. However, even current operate on the principle harm, reducing virus’s ability to grow. Reduced growth has undesired side-effect host immune response below that infection wild-type. Might some instead lead an increased response? We use mathematical models dynamics virus innate and adaptive immunity explore tradeoff between pathology immunity. find modification immune-evasion pathways indeed reduce yet enhance Thus, vaccines can, in principle, directed safe create better than is elicited wild-type virus.

Yersinia pestis with regulated delayed attenuation as a vaccine candidate to induce protective immunity against plague.

Two mutant strains of Yersinia pestis KIM5+, a Deltacrp mutant and a mutant with arabinose-dependent regulated delayed-shutoff crp expression (araC P(BAD) crp), were constructed, characterized in vitro, and evaluated for virulence, immunogenicity, and protective efficacy in mice. Both strains were highly attenuated by the subcutaneous (s.c.) route. The 50% lethal doses (LD(50)s) of the Deltacrp...

Modulation of Innate Immune Mechanisms to Enhance Leishmania Vaccine-Induced Immunity: Role of Coinhibitory Molecules

No licensed human vaccines are currently available against any parasitic disease including leishmaniasis. Several antileishmanial vaccine formulations have been tested in various animal models, including genetically modified live-attenuated parasite vaccines. Experimental infection studies have shown that Leishmania parasites utilize a broad range of strategies to undermine effector properties ...

HIV-1 Gag-specific immunity induced by a lentivector-based vaccine directed to dendritic cells.

Lentivectors (LVs) have attracted considerable interest for their potential as a vaccine delivery vehicle. In this study, we evaluate in mice a dendritic cell (DC)-directed LV system encoding the Gag protein of human immunodeficiency virus (HIV) (LV-Gag) as a potential vaccine for inducing an anti-HIV immune response. The DC-directed specificity is achieved through pseudotyping the vector with ...

Sculpting humoral immunity through dengue vaccination to enhance protective immunity

Dengue viruses (DENV) are the most important mosquito transmitted viral pathogens infecting humans. DENV infection produces a spectrum of disease, most commonly causing a self-limiting flu-like illness known as dengue fever; yet with increased frequency, manifesting as life-threatening dengue hemorrhagic fever (DHF). Waning cross-protective immunity from any of the four dengue serotypes may enh...
